We have mice injected w/ specific cancer cell lines. The cancer cell lines failed to pick up the luciferase bioluminescent tag. We have a rubric of categories to measure the mice, but I was curious to see if there were any other variables that we could measure that would give proxy measurement for changes in tumor size.

In vivo studies FVB and 129-E mice were purchased from Charles River Laboratories. Tumor cells were implanted in 7-9-week-old mice subcutaneously at optimal doses as indicated in figure legends. Tumor length and width were measured by caliper and the volume calculated by the equation: 0.4*length*width^2. For bioluminescence studies, images were obtained with the IVIS Spectrum imager after intraperitoneal (IP) injection of D-luciferin (150mg/kg). we initially wanted to use ivis imaging but our cells lost the luciferase gene. thus we have no bioluminescence.
do you know how to practically measure these tumor sizes with the calipers? Are the mice first sacrificed and the tumors excised before measurement and application to equation?
Is there anyway to continuously measure tumor size in vivo without sacrificing the mice?
