Hello!
I have been having the following blot after western blot transfer, and I am not sure what the problem is...
I make a resolving and stacking gel following the TGX fastest 12% acrylamide kit from Biorad, with fresh 10% APS and TEMED.
I run my samples for 70V at first, and then 100V after the migration front is 1 inch past the wells.
The migration front is always wavy...
The ladder on the gel looks fine (straight).
I then transfer at 100V for 45 mins using a PVDF membrane pre-activated with 100% ethanol and fresh cold transfer buffer.
The photo is the blot after the transfer, stained with Ponceau S.
Please any help or advice would be appreciated!

maybe there are too many salt in your sample, try to dilute your sample and run another gel
