Every time I inject a sample, I weigh the vial before and after. When I do a 100 uL injection, about 100 mg should be injected. Usually, little to no sample is injected.
At the time of injection, the pressure falls from 3000 psi to around 100 psi and then rebounds quickly. Perhaps it is injecting air?
I have tried purging the injector many times, but this does not help. I cannot see any bubbles in the syringe.

According to your information, needle/barrel of syringe/injector port has been blocked. First, clean it and then we will see.

Hi Jeff, this increase in pressure in your 2695 Alliance could be injector valvule clog. You should try clean this, I belive in your HPLC menu has one step for clean up injector.

Check whether your syringe is leak proof and clean. Purge the whole system and equilibrate or condition it before injecting any sample. Inject air sample and check for the backpressure variation.
