Isolated ovaries can be kept in culture in two ways depending on the age of the donor. If the ovary is from a newborn animal (rodent) it can be kept in culture using a floating disc. There are many publications using this method but one example is described in Tuttle et al., 2009 Hum Reprod. 6:1452-9. Now, if you are trying to study functional characteristics of the whole ovary in culture then you are forced to use a much more complicated method of whole ovary perfusion and maintenance in culture. For details of the method see Jarrell et al. 1993 Reproductive Toxicology 7 Suppl 1:63-8. This is a review paper that will lead you to several of Dr. Jarrell's publications that detail the methods and provide insight into useful outcome measures.
