I didn't quite understand your question, but for the choice of bacteria, it will depend on whether your plasmid is replicative or not in its new host cell.

Thanks, Sofiane Benyamina for the response. What I mean to say is can we transform E.coli with plasmids of other bacteria and check the changes in it? For example, suppose there is an antimicrobial-resistant gene in the plasmid of Klebsiella pneumonia. If I isolate that plasmid, remove that gene, and then to check whether antimicrobial resistance is lost or not, do I need to transform it into a competent K. pneumonia bacteria, or I can do that in E.coli also?

But yes, I agree with your point that we need to know whether the plasmid of other bacteria is replicative or not in e.coli.

if the plasmid it is replicative in E.coli and E.coli does not have this resistance gene on its chromosome, I don't see why you can't check this in another bacterial, unless there is specificity expression of this gene relative to the bacterial host cell.

The answer is that it will depend upon the plasmid. Many plasmids are broad host range and will replicate in numerous hosts, many other plasmids are not and will only replicate in limited number of hosts. So it very much depends upon the plasmid and the host it is from. Generally if the natural host is an enteric bacteria, like Klebsiella to use your example, it probably will replicate in E. coli. But for example from Pseudomonas, then it may not.

Do note that multiple drug resistance plasmids are often broad host range, catabolic or degradative plasmids are often not. But there is no universal answer.
