I am trying to transfect MDA 231 cells but my efficiency is too low. I tried up to 2ug plasmid (7.5KB) in 12 well plate with different transfection reagents (PEI and Lipofectamine 3000)
For PEI i used 2 ug of plasmid: 6 ug of PEI
For lipofectamine 3000 i used 2 ug of plasmid : 2ul reagent :4ul of p3000.
The resulted efficiency was
MDA-MB-231 is known to be a bit tougher than the other MDA-MB cell lines in terms of transfection. Using a targeting reagent (https://altogen.com/product/mda-mb-transfection-reagent-breast-adenocarcinoma/) can help you get higher efficiencies (siRNA transfection should be around 80%, while plasmids will be a bit lower). 10% transfection efficiency means that you have a problem with your reagents, and you should also check to see if your selection marker is accurate.
Check your cells for mycoplasma - you should have better efficacy than that.
For Lipo3k https://www.thermofisher.com/uk/en/home/references/protocols/cell-culture/transfection-protocol/mda-mb-231-cells-protocol.html recommends 1 ug w 2ul reagent and 4 ul Lipo
