Hello,

I recently started culturing mouse primary aortic smooth muscle cells (purchased from Cell Biologics) with the media supplied by the same company (5% FBS and a whole lot of other stuff).

I've expanded the cells and they're at P4 right now, and their morphology seems to be all over the places. At this stage, is it normal for cells to be a mixture of spindled-shaped, large & rounded, and cobblestone-like?

Also, since they vary in size and shape and grow in clusters, it's almost impossible to count cells accurately under microscope. Are there any tips on counting them? Or a way to assume how many cells there are when a 100mm dish is full?
