MEM or DMEM media supplemented with 10% FBS; 0.1% Antibiotic solution; L-Glutamine and 1mM Sodium Pyruvate should just be fine for glioblastoma cells like U87MG or SH-SY cells. By the way which cell line are you working with?

I am working with human glioblastoma cells

for my cells, I have been using DMEM/F12

but I noticed that after 2/3 days my glioblastoma cells are starting to dye and I don't know why?

With humen glioblastoma cells you mean primary, patient-derived cells?

For that I'm using the following medium composition quite succesfully:

480 mL DMEM/F-12+GlutaMAX.

5 mL amphotericin B (250 μg/mL).

500 μL gentamicin (50 mg/mL)

2.5 mL HEPES Buffer (1 M).

10 mL B27 Supplement minus vitamin A

supplemented with EGF and bFGF to a final concentration of 20 ng/mL

and culture the cells as spheres on Greiner petridishes (Cat-No. 633161).
