Dear Bhagya, I would suggest that you keep serum frozen at -80°C if it is for a long period of time (e.g. several months or more). Regarding lyophilized serum a temperature of 4°C should be fine, as long as the residual moisture content is low (typically less than 2% by Karl-Fisher assay). If the residual moisture content is not low enough there will be some loss of activity of labile components, and the reconstitution with the solvent (e.g. water) may not be complete (insolubles, particles). I may suggest some thing more about the serum storing. It is always better to make aliquot of 200-300 micro litre of your serum sample and lypholized completely. and keep all the vials at 4 degree temperature. When ever require you can take a vial and reconstitute and use. Repeated freezing and thawing of a single vial may problem for degradation of protein and loss of activity.
