Soft agar vs Matrigel better to look at colony formation within normal tissue?

Hi, I am working with oesophageal cells and treating them to induce stemness. I would like to test for proliferation and colony forming ability. Would a soft agar assay or matrigel be better to investigate colony formation? I know that soft agar assay is typically used in cancer cell lines but can it also be used for stem cells?
