I am doing CRISPR/Cas9 transfection in SW620 and HT29 cell lines using Lipo2000. I have got transfection efficiency of 30-40% in SW620 and less than 30% in HT29.

Thanks for answer. What is your transfection protocol using Lipo2000? I am testing RNAiMAX and Lipo3000 using 7.5, 10, 15, 30 ul in 6 well plate.

I had some experience in HT29 transfection. these cells are very difficult to transfect and they do not transfect well or react well to lipofictamine

I have not tried SiRNA transfection in these cells. but they do not transfect well with plasmids
