Hi all,
I have been trying let B16F10 cells to produce and secrete Melanin upon a-MSH induction. Cells were induced with 50nM, 100nM and 500nM a-MSH in complete DMEM including 0.5mM L-tyrosine without phenol red for 6 days. In the end of 6 days, there wasn’t change in the media Color (brown-black). Cells passage number is just 11 and the cell pellet was brown before seeding for the experiment. What would be the issue? I need to relatively measure extracellular and intracellular
melanin production. if you point the problem and/or share your protocol, I would be glad.
