if i want to detect Prpsc through WB, then i need to make brain homogenate and sonicate it first and centrifuged at 10K x g, but whether i will put PK after centrifugation or before? i personally think after centrifugation only supernatant needs to be given Pk and incubate in 37 degrees for an hour...am i right?
