I have been using COS1 and BHK-21 cell cultures. I coat the coverglasses with Poly-DL-ornithine (Sigma). Cell attachment is poor for BHK-21 and fair for COS1.
Now I am moving to Poly-l-lysine + laminin and gelatin coating.
Some protocols say to dry after coating and some not to dry.
Which is better? Thanks for sharing your experience and suggestions.
In my experience, drying is ok. I think it may increase the matrix attachment to your coverslip. However i never tried without drying or with the combination you wil perform.

Thanks Benjamin. My neighbors here coat with poly-l-lysine overnight and then with laminin overnight and without drying. They don't have any problem with their cells (they use for stem cells).
For attachment of endothelial cells (HUVEC/HPAEC) I have used either gelatin or poly-L –lysine. I have observed that after coating if I dry the slide/plate then it will be hard to harvest the cells by trypsin-EDTA treatment. Since longer time treatment with trypsin-EDTA may not be healthy for the cells I recommend not to dry the plates/slides after treatment with the adhesive agents.

I agree with Tapan, I used poly-Lysine and Laminin coating for rat neuronal cells culture and I used not to dry the coverslips. You can perform the coating overnight at 4°C and wash your coverslips just before seeding cells.
