I recently performed a competitive ELISA for nAbs to SARS-CoV-2. Samples values were over ranged, so they need dilution. Since the diluent from the company is not enough for that (Wantai SARS-CoV-2 nAbs, ELISA) I wonder if I could just dilute my samples with sterilzed PBS?

To prevent non-specific interactions and adhesion to the plastic surface, sera diluent should also contain an amount of non-human proteins (other mammalian) and a non-ionic detergent (Tween 20). For a 100 ml house-made recipe diluent: add 1g of BSA (bovine serum albumin) plus 50µl of Tween 20 to 99ml of PBS and sieve through a 0.45µm syringe filter.

I agree with Aissa. You should use protein containing buffer.
PBS with 1%BSA and 0.05%Tween-20 would be a better choice.

Christina-Nefeli Kontandreopoulou Try to imitate the original diluent. If this should not be possible, PBST with BSA is a good choice. But you need to validate the buffer / sample dilution by applying it to the standards, too (at least in a test setting), otherwise you might run into trouble if someone is challenging your test (the results probably will send some people into quarantine and they might not like that).
