I serum starved my smooth muscle cells ( three 6 well plates) yesterday, and I was planning to stimulate these cells with drugs in following day, However, when I get to do my experiment in the following day, I did not find the control drug as someone has used it, so my question is what should I do in this case? should I add serum to these serum starved plates and wait till I find an aliquot of the drug next Monday ? because if I keep them without serum they will die.

. Please do not continue with those cells as starving your culture of serum for too long will also lead to reduced cell survival and increased apoptosis. Serum starvation could induce unique stress signaling pathways that could have an effect on the results later on when you use these cells for bioassays.It also depends on the cell type. Some cells can tolerate 24 hours serum starvation while there are other cell types which may be sensitive to serum starvation. You never know what may be going on inside the cells. You would always want your cells to be normal and healthy for invitro bioassays so that you may obtain accurate results.
The next time when you serum starve your cells, you may do so in a medium containing low serum concentration (relative to normal growth medium), typically 0.1–0.5%, but sometimes as high as 1-2% or as low as 0.05%. Low serum concentration in the culture medium will help cells to remain healthy but not proliferate.
