Can anyone tell me what is the seeding density for MC3T3-E1 cells per cm2? How many cells should be seeded in a 12-well plate? As general rule for a 12-well plate I found a seeding density of 0.1 x 10^6 (thermofisher; useful numbers for cell culture). Does that work for MC3T3_E1 cells?

Actually the seeding density depends on your purpose. If you will perform cytotoxicty test it is too much but if you will perform cell migration assay it can be better. Also MC3T3-E1 does not like over confluency in general at late passages. So you should decide it regarding to your main purpose.

For 12 well plate I recommend maximum 5x10^4 cells. As I emphasized before it should depends on your experiment.
