I was told my low RNA-concentrations extracted from a pigs stomach could be due to the amount of RNases in the stomach. I was not cut up at the slaughterhouse, so it contained all the stomach contents on the way to the lab. I cant find it in the litterature or on google. Can someone help? I need a reference for my assignment.
Please do not flood ResearchGate with repeated queries. The responses other researchers gave in the your earlier posts (on the same topic) may be helpful and the references they provided might give clues and how to proceed further in your search for an explanation. Furthermore, do not get fixated with the hypothesis that was first given to you. It is much easier to search for literature nowadays and get full-text copies of potential articles that may shed light on your concerns.
The hypothesis on "the amount of RNases in the stomach," I would argue, is not entirely helpful. First, there are RNases everywhere. This is why RNA-related experiments should be conducted with instruments, reagents, equipment and containers that are RNase-free. For one, please examine several points in workflow that may explain your findings. Were your protocol and extraction kit, if applicable, optimized for RNA extraction from a sample as heterogeneous or unique as a pig's stomach? What regions of the gross tissue/organ did you conduct the sampling? Did you try sampling for RNA on an intact stomach sample or on a homogenized one? Is your technique for measuring the RNA content of your samples sound? If the RNases in the pig stomach is really that substantial, what interventions could you have done to inhibit them?
