the max volume you should inject would be 1 ul...some studies used 5 ul which I think is too much. the total hemolymph volume is about 12-15 ul per bee. insects have open circulatory system, so anything you inject into the body cavity will be into the blood. I normally inject into the inter-segmental membrane. but for some studies I inject into the thorax.
I know about the open system and the iRNA studies. I was only wondering if it is a good way to make dyes diffuse until the CNS. For live two-photon microscopy we usually add a drop of dye on the exposed brain, but of course the deeper layers of neurons are less stained due to incomplete penetration. That is why I was thinking at the injection as a way of staining everything in a more homogeneous way.
