I know how to isolate HUVECs from umbilical cord quite well, but
I want to ask about use of syringe filter during huvec isolation.
It seems that I have used syringe filter with 0.22um filter unit during
isolation of HUVECs,
but i found that 0.45um syringe filter is suggested in my related protocol.
I want to know which one is optimal, or whether i can use any type
of syringe filter i want during isolation of HUVECs.

You are using the filter for what reason? If just to filter the buffers for sterilization purpose that Ok but if you put the cells through 0.22 µm filter you will get nothing because the size of the filter pores are too small for the cells.
Hi!You don't need to use filters because (assuming that you use collagenase for isolation) HUVEC are the cells that you will isolate from the vein of the umbilical cord.
