Does anyone have a protocol for performing morphology staining of monolayer cells in culture? For cell cultures- are Giemsa staining or H&E staining the best options? I only want to visualize cell morphology of adherent cells.
I use to do crystal violet staining for watching morphology of adherent cells. It works well. I don’t have the protocol now but I can get if you need it.
This is in no way an optimized protocol but I have done H&E on myoblasts to visualize their morphology (just wanted to confirm something).
I am assuming you already have the cells on a slide.
- Hydration, 1-2 dips
- Hematoxylin for 2min
- Wash in running tapwater for 2 min
- Eosin for 2-3 dips.
- Dehydrate, 1-2 dips
- Xylene and Mount with DPX.
The cell retained morphology, no lysis was seen and was able visualize the cells.
