Do anyone have experience with the alamar blue assay monitoring cell viability for fish cells, or cells incubated at lower temperatures than 37 degrees C? I am working with cells incubating in temperatures such as 20 and 28 degrees C. Is 37 degrees necessary for reduction from blue to red color?
Also, how do you calculate a 96 well plate? Is there a easy way to calculate this without having to calculate each well/sample?

There isn't many differences between working with mammalian or fish based cell models for viability assessment. What the temperature might influence is the incubation time and the number of cells necessary per well. I added this protocol that I usually follow to establish the cell number and time for new models. I usually incubate for 4h at 25ºC, but addapt the number of cells depending on the cell line.
