Hello I-Lun Hsin, If you can not see any change in their relative cell cycle distribution - does that include hyperdiploid cells(https://www.nature.com/articles/nprot.2006.238.pdf)? If no evidence of increased apoptosis(hyperdiploid peak and/or annexinV expression) - you could run parallel cultures with/without drug and do comparative daily counts and viability assessment - PI or trypan blue by microscopy, to determine if the cell cycle had been inhibited. Further, you could label cells with a cell tracer eg CFSE, ef450 and track division times and population dynamics. Best wishes
