Dear Researchers,

I use Matk, psb and rbcl as universal primers for PCR and I have to use them on number of DNA samples of different plants.

So, I have to first run touchdown program to know the amplification status.

Then I have to run gradient, which isn't economical.

So, can a primer set have more than one annealing temperatures? or each primer set has a specific annealing temperature always?
