I'd appreciate any input on the advantages/disadvantages of using a positive or negative T cell isolation kits (STEMCELL EasySep) for in vitro assays.

I would like to isolate T cells from human PBMCs and culture them under various conditions for T cell proliferation assays and cytokine production.

Should I use T cells that have been negatively selected (without any attached magnetic beads) for in vitro assays? I'm wondering if the beads from a positive selection kit (CD3+) may interfere with T cell functionality...?
