I am working on HSV-2 and dsDNA induced autophagy on patients PBMCs, and I would like to use a potent autophagy inducers (mTOR inhibitors and autophagosome formation inducers) because I am having troubles inducing autophagy following dsDNA transfection and virus infection. Any suggestions? Does anyone has experience with Rapamycin or Everolimus? MG-132 or SAHA?
All comments/ideas will be appreciated.

Serum withdrawal for 3-6 hours or the treatment with rapamycin (100nM for 3-6 hours) can be the positive control of autophagy induction. You can easily check the autophagy activation with the formation of GFP-LC3 puncta under microscopy or performing WB of LC-3 II and p62/SQSTM1. To rule out the influence of autophagy flux, you can combine the above mentioned treatment with CQ or bafilomycin A1.
