Hi Mamatha, I would not recommend using whole blood as a growth media. First the blood has EDTA which is toxic to cells and you will have RBC hemolysis that releases toxic heme groups.
Whole blood may contain a multitude of proteins, ranging from growth factors, which may not be specific to your cell's needs, to proteases which will for sure degrade your cells. Depending on you're goal you should use serum, or platelets. Or best would be to extract the proteins from blood and add them to a culture media, if you want to see what effect blood proteins might have on cellular growth. (Don't forget to add protease inhibitors.)
