You may obtain 300 to 400 000 cells per well depending on your strain. Be careful to passage the cells before full confluency to keep them proliferating.
Hi Purva - the amount of cells you will obtain will depend upon the passage level or cPD (cumulative Population Doubling) of your MSC and your time of harvest.
For example "young" MSC (low cPD or early passage) may have a doubling time of 2-3 days where as "old" MSC (cPD ~25) will have a much longer (population) doubling time (and eventually will stop growing altogether).
Also as MSC reach confluence they do become contact inhibited - I've found that MSC (cPD ~12) seeded at 10 000 cells/cm2 in a T75 give almost the same number of cells 5 days post seeding as they do at day 7 (and they were still sub-confluent at day 7 at ~ 80-90%).
Similarly the type of media will also affect the yield at the end of your experiment - the quality of the FBS, presence of growth factors (bFGF) etc.
Thus your question is a little difficult to answer without more information.
i.e. how old (cPD) are your cells? How long are they in culture for? What confluence level are you harvesting at?
