Has anyone used phosphatase inhibitors on live cells? My goal is to stimulate the samples and add phosphatase inhibitors to prevent dephosphorylation while surface staining step. The problem is that my surface antibodies look bizarre if I fix first and then stain. Instead, I am hoping to block dephosphorylation using phosphatase inhibitors, surface stain, fix, permeabilize and then IC stain.

I am not sure if the phosphatase inhibitors like Orthovanadate or NaF are cell permeable.

Anybody has any ideas/suggestions with that?
