I am performing PDX ALL in NSGS mice, but something curious happens: some leukemias develop very well , but after a time the hCD45+ cells show a huge regression while the mCD45+ cells turn back to growth. Do you beleave that the issue could be the NSGS mice lineage? Do you have ever seen it already? It is like a spontaneous regression of leukemia.
I'll be really thankful if you give me a feedback to help me undesrtand what is going on with my PDX, please.

I worked with aml, not all, so i don't know if i could be helpful. In my experiencie not all the aml patients transplanted develop leukemia phenotype in the mice. A possible explanation is obvius technical, not enough cell, or not well injected, or directly not engraft. In some cases the first weeks or inclusive months are detectable, but eventually regress. In my opinión, could be for two reasons, pretty related. Firstly, some of the patients used were not isolated the cáncer stem cells and or tumor initiator cells, are malignant but can't repopulate a new tumor niche. The other reason, is environmental dependency, sometimes microniche and supporter/inmune cells from the patients are reprogrammed for the own cáncer cells and cáncer cells generate dependency. Maybe transplanting whole bone marrow cells from patients could solve this problem, although is a more dirty model. Good luck.
