As a final project for a college course, we need to do a primary culture of non parenquimatic liver cells from mice. In our first try we obtained a great amount of white blood cells instead of hepatic cells. How can we decrease the amount of white blood cells obtained from a murine hepatocyte extraction?
To ensure effective liver perfusion during the extraction procedure, it is vital to eliminate a substantial number of blood cells. This can be achieved by using a cannula to flush the liver with an appropriate buffer solution, typically a heparin-supplemented HBSS solution. By doing so, blood cells present in the liver vasculature can be efficiently removed.
Additionally, it is important to adjust the digestion parameters in order to minimize the release of leukocytes. This can be accomplished by optimizing various factors such as enzyme concentration, digestion time, and temperature. One effective approach is to reduce the concentration of collagenase or other enzymes used for digestion. This adjustment helps minimize tissue disruption in the liver and ultimately reduces the contamination of leukocytes.
