Oocyte swelling assay detail protocol. Aquaporin inhibition studies. Any suggestions for other studies?

I am completely unaware of swelling assay as i am from a different back ground. I have a compound which i hypothesize that it will inhibit aquaporin. I am not sure that it would make any gene or protein changes in expression level. When i read through all the literature every one has proved its inhibition with swelling assay. Please do suggest me if there are any alternatives and if not please do provide me with the detailed protocol for the same.
