I am seeing non-motile black dots of almost equal size along the periphery of skin melanoma cell line A375. They are present in almost all the cells. They don't turn the media turbid. However when cultured for a long time cells gradually change their morphology towards a spherical shape. They are negative in DAPI staining and are not eliminated even after pen-strep treatment. I am attaching an image of the cells here. Any kind of suggestion on how to get rid of them will be very helpful. Thanks in advance.
They are probably just cellular debris from dead cells, or even debris from the living cells. This is kind of normal, but check the pH of the culture medium. You can only consider contamination if you see the dots multiplying day-to-day and the medium turns turbid and yellow. You can try to get rid of it with some washes using PBSA solution next time you are changing the medium.
