I have an experiment where I am trying to see if IL-6 alters the expression of some enzymes. In order to test that I used a neutralizing antibody for IL-6 and an Isotype control. After doing the experiment and performing some rtPCR to measure the expression of the enzymes of interest it seems that in some cases the isotype control by itself has an effect in the expression of the enzymes when compared to normal saline controls. It was a large experiment with 9 groups and now I am trying to figure out what to do with it. Because if my isotypes had an effect by itself, whatever else I see with the IL6 neutralizing antibody can still be taking into consideration?
