Anyone knows if monocites proliferate when in culture with M-CSF, N2 and serum? I mean create a clonal expansion in vitro, not only diferenciate into macrophages? Bibliografy is some confusing, and my experience is limited.
I found this article to be helpful. I have not done any work with monocytes but I have had to do a small literature research on it. The article below is an investigation around the means better suitable for culture and proliferation of CD14+ monocytes. It is a starting point.
Once you isolate monocytes, you could plate them with no cytokines/growth factors and use them within 24-48 hours. The longer you keep them, the more they will die. Once you plate monocytes with M-CSF, they will start differentiating into macrophages (and a mix of macrophages and dendritic cells if using GM-CSF). So long story short, monocytes typically don’t expand in culture; they differentiate to other cells types and proliferate based on what you add to the culture.
monocytes do not proliferate in culture in vitro. Once you isolate them, you can differentiate them into M1-type macrophages, M2-type macrophages or dendritic cells in vitro.
Alternatively, you can work with immortalized cell lines of monocytes/macrophages that are able to proliferate (THP-1 cells in human or RAW264.7 cells in mice for example).
