We have a 96-well assay we use to measure mitochondrial membrane potential. A 200 uL volume of cell resuspension is in each well. We want to add a membrane uncoupler, like FCCP, to uncouple the mitochondrial membrane potential. Howeve,r because of FCCP's solubility in DMSO, we can only add 0.2 uL of FCCP to maintain a 0.1% final concentration of DMSO. 0Does anyone have suggestions for alternative ways to chemically uncouple the membranes? We want to be able to treat with a larger volume than 0.2 uL (like 1-2 uL). Thanks!
