for callus cultuer just do one thing try 2-4,d 05,1.00, 1.50, 2.00 and 2.50 mg /l alone or in combination with 0.50 mg/l kinetin. it is prerequiesite that for callus induction keep your cultures in dark for about one month. and see the results.

for meristem culture use axillary bud as an sourse of explant material use nodal explant and from nodal explant you will get multiple shoots in MS medium supplemented with 0.5, 1.00, 1.50,2.00 upto 5.00 mg/l (BA and or Kinetin) and if TDZ or 2-ip than you have to use 0.10, 0.20, 0.30, 0.40, 0.50 etc for axillary shoot proliferation from nodal explant. best of luck dear if you need any litrature than feel free to contact me.
