I am expressing an mCherry tagged protein. Yield of total protein is good but the fluorescence intensity is very low (ie. Protein assays and gels show a lot of protein but this does not correspond to fluorescence, leading me to believe the tag itself is unideal).

Has anyone experienced this? We have tried changing lots of variables like time, temperature, IPTG and OD at induction.

We have had other mCherry tagged proteins that worked well but this one seems to be a fussy one...
