My MCF10A, and MCF12A cells as well, are growing exteremly rapidly. Even if I plate them at less than 10,000 cells/cm2 in 75cm2 flask, within 48 hours or so the flasks are completely confluent. This was not always the case, it used to be that the cells would take about 5-6 days to become confluent. I have tried using different basal media formulations, decresing the levels of horse serum from 5% to 2.5%, I have used less L-glutamine, less sodium pyruvate, and have still not been able to get them to slow back down. I always followed the media prep as outlined here: 50:50 DMEM/Hams F12, with, 2.5mM L-glutamine, 15mM HEPES, 0.5mM sodium pyruvate, 1.2g/L sodium bicarbonate, 20ng/ml human Epidermal Growth Factor (hEGF), 100ng/ml cholera toxin, 10µg/ml bovine insulin, 500ng/ml hydrocortisone, and 5% horse serum; grown at 5% CO2, 37C, and 100% humidity. I do not use any antibiotics when culturing my cells unless there is a reason to do so, and only for a very short time, which is very rare anyways. The morphology looks the same, but the growth rate is just incredibly rapid.
Does anyone have any suggestions as to how to slow down the growth of these cells. It seemed like it started happening for both the MCF10A and MCF12A cells all of a sudden one day?
