Does anyone know of any differences between the HK1.4 and the Al-21 clones, as far their ability to distinguish cell populations by Flow cytometry. I understand they are of different isotypes, but is one more preferable to the other?
We tested both clones and had very similar results, at least for the monocytes/macrophages. As long as you are using the same clone throughout the experiment - you can choose any.
If you're treating with the anti-Gr-1 antibody RB6-8C5, it may matter. In the figure legend of figure 5 in the listed reference, it notes that clone AL-21 binds to a different epitope of Ly-6C than the anti-Gr-1 antibody. I don't know about HK1.4's binding epitope though. I would try both if this applies to you.
