So we have an FPLC system which is the P900 that has been acting wierd for a week or two. Even though no air should be in the system the pump introduces air anyways and makes the clicking noise that a pump does when it has air in it. And i have tried to suck all the air out with a syringe at a valve right after the pump which is how you do it if you accidentally got sir in the tubes. But this time the machine gets the air in so this doesn't work. And i can't find any leaks anywhere so i don't understand how this is happening.
Has anyone else have had a similar problem and how did you solve it?
A few basic areas to check:
Most likely the inlet valves are stuck. The valves can be removed and cleaned in an ultrasonic bath if you cannot clean them in place. Don't disassemble the valve; just put the whole valve in the cleaning solution and into the ultrasonic bath. The manual recommends to use 100% Methanol - I have had good results using a surface active detergent. In both cases you should repeat with water.
