Need a protocol for sonicating the Hep3B cells using Ultra-sonication Instrument.

Liquid medium allows Ultrasound waves to move through and break down the cells. It is not clear to me why you want to sonicate cells without buffer. Can you explain your equation more?
Ahmed E. Dhamad Yes Ahmed, I want to test an enzyme activity in cancer cells. so that i am using a commercial assay kit. The troubleshoot i get here is that I can't standardize the amplitude and pulse break in ultra-sonic instrument. So only i'm asking for a protocol which includes the - Amplitude % , Pulse on and off time. If you know then please send me it.
The amplitude% depends, generally, on your sample volume and a cell type. I used 10% Amp. for 50-200 ul sample V for 293T cells. Pulse, I often use 5-10 times with 10 second on/off. Hope, that answers what you have asked for.
