My Hek293 cells are grown in MEM media +10% FBS + 2mM glutamine.
When the container the cells are in is moved, a clusty of cells would collapse upon itself. I know its not the quality of the FBS, flasks, or cells themselves because other cell lines in my lab are growing fine. And I have thawed multiple vials from liquid nitrogen and I get the same result.
Your help would be greatly appreciated
You can tell a lot about the health of Hek293 by their morphology, can you post a picture?
In general, Hek293 are quite weakly adherent but stick alright to tissue culture-treated flasks so long as they are not "sloshed around" too much.
Maybe your cells were not in a perfect shape before freezing (HEK293T cells are very sensitive and will not fully recover if left in the culture too long between 2 splittings).
Also, try using DMEM instead of MEM.
And split them regularly, maybe they will recover.
The cells should be fine with EMEM. In fact, that's what ATCC specifically recommends and I have used it with great results for quite some time.
