Image analysis of the bands is a relative measurement, a kind of semi-quantitation. It is reliable to compare in two and more bands. Protein bands are more close to real in relative terms than the amplified RT-PCR DNA bands, but not unamplified DNA fragments. This comparison is enough to estimate the amounts. This could be demonstrated by loading the known amounts of DNA and proteins together.
