Media for freezing of cells for culture.
Requirements
700ml RPMI (2mM L-Glutamine) 200ml FCS 100ml DMSO
Method
- Filter sterilize 700ml of RPMI-1640 (2mM L-Glutamine) and 200ml fetal calf serum (FCS) through a 0.22µm filter
- Add 100ml sterile dimethyl sulfoxide (DMSO)
Do not filter DMSO, it will dissolve the cellulose acetate membrane.
- Aliquot into tubes as appropriate
Store the tubes at -80oC for up to one year.
generally the medium which you use to grow and culture your cells is the one used that is for freezing unless your cells are something special and the manufacturer has given some special formulation.
what are the cells that you have?
I agreed with the answer above. i usually grow culture cells in Dmeme and always use DMeM, FBS and DMSO for freezing cells. Another way is to freeze only FBS and DMSO.
I want to add a recommendation, never leave your cell at -80°C, the last sentence "Store the tubes at -80oC for up to one year " it is wrong. You freeze the cells at -80°C in correct freezing container for few days and then store in liquid nitrogen
