I have obtained from a friend lab A549 cells (human lung carcinoma) and after growing them for a short while their morphology seemed to me somehow strange (see picture) compared to some old A549 cells we used to have and unfortunately lost!! Does the morphology in the picture seem normal to any of you? Has anyonne seen the cells like this before? The cells are cultured in high glucose DMEM growth medium suplemmented with 10% FBS, Glutamin and antibiotic/antimytotic (Pen/strep).
Hi, I also work with A-549 cells and we grow them in High Glucose DMEM supplemented with 10% FBS and 1X Ab-Am solution. To me, the morphology looks somewhat elongated. Have you passaged it 2-3 times before you got this image?
I noticed that and this is the reason I am asking here!! The morphology looks more like fibroblasts to me rather than epithelial cells!!
To reply to your question, yes I did passage them 2-3 times.
Yeah, they look like fibroblasts or mesencymal cells now. Did you grow them to confluency during one of those passages? I noticed this with my breast cells, that if they got super packed, then I passaged them, their morphology would change (as well as their growth rates). Just a thought. Also as a side note, the cells never returned to normal morphology.
When I first got the cells, the confluency was at about 20%. The cells had similar morphology to the "normal" one, and one day after they obtained this morphology. Never got too confluent. I know they don't turn the other way around as this is my second attempt to culture them. The same issue was noticed in the lab I got them from. To me it is rather a mixup with some other cell line.

Did you grow this in glass bottom dish without collagen coating?
No I didn't, these cells were cultured in plastic 10cm dish from Sarstedt ideal for cell culture!!
These could be either a post-high confluency growth morphology, then their shapes deformed, or that there is a suspicious of these cell line origin. Simply get rid of them and grow another patch from different laboratory; A549 are everywhere could be find.
