We are assaying E. coli cultures for the detection of enzymatic activity in a colorimetric broth under different conditions. But our controls give positive results too early due to, we assume, spontaneous lysis of the bacterial cells during the growth. Does anybody know if there is any additive (eg glycerol, agar, etc) that could be added to the medium in order to reduce this spontaneous lysis of bacterial cells?
