I am working with various strains of E. coli for protein expression by cloning. Recently, I observed that the colonies of E. coli C43(DE3) is different from C41(DE3). The C41(DE3) colonies are archetypal BL21(DE3) colonies with transparent and diffused appearance, while C43(DE3) colonies are compact, smaller and whitish.
Please forward your valuable suggestions on this.

Actually I am using E. coli C41 and C43 for cloning. Since, both the strains are derived from Bl21(DE3), I considered them to appear similar on LB plate. But to my surprise, C43(DE3) was compact with whitish appearance, and C41(DE3) is relatively transparent.
Any given E. coli strain will frequently segregate mutants with altered morphology. Mucoid variants are very common and can be observed as transparent oily larger spreading colonies. This can happen with any strain so it not really something different between C41 or C43 per se, but just the stock you have in your lab.
