Please address the best way to drop a plasmid.
Background: I have a "bait" plasmid resistant to kanamycin and a "prey" plasmid resistant to carbenicillin. After many rounds of streaking on LBcarb and performing a tranformation, I still have colonies that grow on LBkan when patching plates.
I've managed to get bait and prey plasmids separated by doing a plasmid prep from the strain with both then transforming with selection for one or the other. I checked the transformants for absence of the second plasmid via re-streaking like you are doing now.

I concur with Amy Klocko recommendation. It is very hard to get a strain to lose a high copy plasmid (whenever I've tried to do I've had to screen hundreds and hundreds of colonies). But retransformation should nicely separate them and only a minority is likely to co-tranform with both.
