Hi Joana, I have OKF6 normal oral cell line. Yes they are quite difficult to grow. You have used the correct media but I think the viability of your cells from the frozen vial itself is less. My recommendations is to procure the cell line from the company directly after you sign the material transfer agreement. Even if you get the cell line from someone, you have to conduct short tandem repeat to determine whether your cell line is really OKF6 or not. Hope it helps.

Gibco provides the supplements for keratinocytes with the media as well that include EGF human recombinant and Bovine Pituitary extract. Did you add these in your media? And you would also need a antifungal and antibiotic to avoid contamination. Usually, with all this taken into account, they grow pretty well. All the best!

Hi! Many thanks for your answer and hope all is well! Indeed we added the supplements from Gibco to the media, EGF and Bovine Pituitary extract, as well as PenStrep but failed to obtain growth :( What we observed was that immediately after thawing the cells were not able to adhere and grow (even after 24h or more).
